The Sperm Freezing Process

The process of sperm freezing is straightforward and required undergoing blood testing as an initial step, primarily to screen for HIV, hepatitis B, and hepatitis C. It involves a few key steps:

    • Most men provide a semen sample through ejaculation in a private, comfortable environment at the clinic or at home with proper guidance. For those unable to ejaculate, surgical sperm extraction directly from the testicles may be performed.

    • To ensure an optimal sample, men are advised to abstain from ejaculation for 2–5 days before collection. If ejaculation occurred too recently, the sperm count might be lower. Conversely, if there has been a prolonged gap since the last ejaculation, many sperm may lack motility.

    • Laboratory specialists evaluate the sample for key parameters such as sperm count, motility, morphology, volume, viscosity and pH levels.". A single ejaculation can contain anywhere from 40 million to 600 million sperm, depending on the volume of the semen and the time elapsed since the previous ejaculation. This step ensures the sample is suitable for freezing.

    • A cryoprotectant is added to the semen to protect sperm cells from damage during the freezing process. Two techniques are commonly used:

      • Slow Freezing: Gradually reduces the temperature over several hours to prevent ice crystal formation.

      • Vitrification: A rapid freezing method that flash-freezes the sperm, preventing ice crystals and preserving cellular integrity.

    • Samples are then stored in liquid nitrogen tanks at -196°C.

    • Thawing sperm is just as crucial as the freezing process, requiring careful handling to allow the cells to regain their normal biological functions while avoiding sudden temperature changes. Cryopreservation protocols typically involve a controlled thawing temperature of 37 degrees Celsius. Although higher temperatures could speed up the process, they are avoided to minimize the risk of cellular damage. When needed, the sample is thawed using controlled methods to restore biological activity. Cryoprotectants are carefully removed, and the sperm is prepared for use in reproductive treatments such IVF or IUI.
